In microglial cultures, capsaicin increased the [Ca(2+)](i) via the TRPV1-mediated influx of extracellular Ca(2+), and the inhibition of L-type Ca(2+) channels with verapamil (5 microM) inhibited the capsaicin-induced increase in [Ca(2+)](i) and in the IL-1beta and IL-6 release. We show for the first time that capsaicin increases [Ca(2+)](i) in microglia by increasing the influx of extracellular Ca(2+) through TRPV1. These results indicate that capsaicin-induced microglial activation and the resultant release of neurotoxic factors may play a role in the neurotoxic effects of TRPV1
