We found that the activation of TRPV1 ion channels by capsaicin stimulated the release of the proinflammatory cytokines IL-1beta and IL-6 in both cultured microglia and neuron-microglia co-cultures. The intracellular Ca(2+) concentrations ([Ca(2+)](i)) increased in microglial cells treated with capsaicin (5 microM), which was blocked by pretreatment with the specific TRPV1 antagonist SB-705,490. The addition of capsaicin did not stimulate the release of IL-1beta or IL-6 in neuron-microglia co-cultures, indicating a specific effect of capsaicin on microglia, and that intracellular Ca(2+) increases are required to induce IL-1beta and IL-6 release in microglia. In microglial cultures, capsaicin increased the [Ca(2+)](i) via the TRPV1-mediated influx of extracellular Ca(2+), and the inhibition of L-type Ca(2+) channels with verapamil (5 microM) inhibited the capsaicin-induced increase in [Ca(2+)](i) and in the IL-1beta and IL-6 release. We show for the first time that capsaicin increases [Ca(2+)](i) in microglia by increasing the influx of extracellular Ca(2+) through TRPV1. These results indicate that capsaicin-induced microglial activation and the resultant release of neurotoxic factors may play a role in the neurotoxic effects of TRPV1
